What keeps you from putting in a 454 round into the cylinder instead of 45 colt into it?

I was at S&W Retail Store yesterday, I got to handle the Gov... beautiful gun. I was told price should be right around 600, 800 with the laser.. it was really sweet and felt really good in the hand..
