It would have to be 357 mag followed by 9mm and 45 APC and 22LR. I cannot match 75Vettes wall of revolvers, but am pround of my starter set.

NOW THAT'S A GREAT CALIBER, Goldstar225. I FAVOR A SIMILAR CALIBER, MINUS THE 41---WHICH IMHO, DOES NOTHING THAT CAN'T BE DONE WITH A DIFFERENT 2 DIGITS, STARTING WITH A 4.....

My choice .327 mag. For accuracy load the .32 S&W wadcutters, for power the .327 mag.

My favorite revolver is .38 Special
My favorite semi's are .45 ACP and .380

LIKE PICKING A FAVORITE KID.
Only owning 6 handgun calibers (at present), the most enjoyable would be 22's & 45acp. The one I shoot most, 38 special.

.44 Magnum is too much of a good thing. Unless you are hand loading.
Or shooting .44 Specials then it's awesome. When shooting if it feels uncomfortable there's something wrong. .357 and .38 Special is the limit and perfectly comfortable to shoot. Although my new SW22 Victory is becoming fun to just shoot and relax with.

In the end you know what the best calibre is?
The one that works best for you. Everyone is different.
Just find what you like and shoot targets to enjoy.

357 is what I fire the most out of revolvers. Although these days less of the 296/H110 flame thrower loads.
In the semi-auto is it the 45 ACP.

What is your favorite handgun caliber?
I load for about 65 Different calibers but my favorites are:
Self defense (handgun) : 45 Auto (.45 Super is a very close second).
Deer and Wild boar: .45 Auto Rim or .45 Colt (but .44 Spl. is a contender as well )
Small Game and plinking: .22
Oddball fun guns: .22 Jet and .256 Winchester
I sure have no kick with anyone's choice - we all have to work out our own.

I came in to post just what you wrote...
.44Magnum is so versatile. I'm very fond of my 3" barreled 629 W/lock delete installed. For street carry .44Specials are more than capable of getting the job done with 200 grain Gold Dots loaded up around 900fps. For woods carry, (and I do carry this in the woods everytime), I stoke it with 300 grain semi-jacketed flat points pushing 1150fps. And I've got 240 grain .44mag loads for everything in between.
.45acp is a close second in my 1911 and CZ 97B
9mm & .38Specials are tied for a close third carried in CZ P-01/SP-01 and vintage S&W "J" frames.
I would never be able to choose just one. And America still being a free country I never would choose just one. But to play the "If I Had To" question; it would undoubtedly be .44Mag/Special...

.38Super is a pretty awesome round especially out of a 4" or greater 1911.

Hmm, probably .44 Special, though it’s not what I shoot the most these days.
In an auto, I do like the .40.
