My vote for the most comfortable are without a doubt the Pachmayr Compacts.

My daily carry (S&W 642) wears Crimson Trace #305. They have a covered back strap that increases the length of pull (noticeably better for my hands) and they are reasonably comfortable for .38 +P's. They ride just fine in my right front pants pockets in a Mika pocket holster.
