The .357 would have been Doc 'jumping on the bandwagon' with the rest of the FBI after the caliber was introduced for penetrating the crude body armor of the day, and the very thick auto bodies of that time, too. Being a former Texas Ranger he favored his engraved Colt SA that would have been ineffective against armor of any kind.
