When is absolutely, positivly has to be on target-.308 win 155gr2900 fps, 10 yards past a 1000 it hits the target! closer than 10 yards-butt stroke it

.22 LR because I can afford to buy it 5000 at a time, .38 spl. because it is the classic revolver round and .45 ACP because it is .45 ACP!

That's difficult to have just one. First has to be .22lr for obvious reasons.
Next I would vote for 9mm. Cheap and plentiful.
For centerfire rifles....probably .30-06.

Looking at my handloading records, it's 30-06 by far. Also rans are 8mm Mauser, 30 Mauser Pistol, 9mm P, 45 ACP, and 38 special.

As much as I like revolvers for reasons too numerous to take up space naming here, I learned to shoot a centerfire pistol using my Grandfather's R-R 1911 .45 auto. Still these many years later, my undisputed "favorite" would be the .45 ACP in a 1911.
Digressing... On the other hand, I get so much enjoyment out of shooting so many different guns that it seems a travesty to ignore them, so I will add these two:
Shotgun - 28 gauge
Centerfire rifle - .222 Remington
If I made up a list of "honorable mentions," it would be a mile long.

9mm Luger, Parabellum, NATO, etc. Get it anywhere in the world, even where you can't find .22LR (second choice).

I know it isn't the sexiest caliber out there but I enjoy shooting .38 Special better than anything else.

Don't know if I could pick between 22LR, 9MM Luger, and 45ACP. I think the 9MM Luger is kind of the middle ground between the 22LR & 45ACP, with affordability and power.

38/357 gets my vote. I love my 44 magnums and specials and the .22 revolvers, but none are as versatile as the 38/357, IMO. From light plinking loads, target work, self defense and light hunting loads in a package that fits my hands best and carries easier than an N-frame, I like the 38/357 and the guns that chamber it.
Thanks to the OP, now I know what gun to take this weekend when I visit relatives in the "country" - model 19. "Everything you need and nothing you don't." I stole that line from somebody.

if it all cost the same i might lean toward bigger is better though lol

.22 and K22 3rd Model 5 screw.
.22 and the Remington Model 12 I’ve yet to buy.

Let me start out by saying there are all sorts of calibers I have not tried that I'd like to.
Lately, I'd have to say .223/5.56. I've never had so little meat loss on a deer, its cheap to shoot, the military surplus is pretty accurate. Magazine are plentiful and cheap for my gun. virtually zero recoil. Whats not to like?

As far as hand gun caliber I shoot the most would be .38/.357 Models 10, 14, 15, 19, 60, 66, 638, 640 & 642. My favorite caliber however is .45 acp in model 625 or 1911.
Favorite rifle round is .22 Hornet with T/C Encore.

The question asks for favorite..not most practical. That is a very different set of calibers.

Semi auto .357 sig It's just a great round with great ballistics

They are not the ones I reach for on an everyday carry basis, but my .41 mags make lots of trips to the range and the woods.
The rifle I reach for most often is the ugliest one I own. A Rem. stainless, mountain rifle, black Teflon coated, scratched so often that it is turning stainless again, inconvenient blind magazine. The caliber is .280 Rem. It goes in the rain, snow, or when I pay to hunt and want something I have confidence in. It is extremely hard on deer.

Don't even have to think about it. The .41Magnum is the king in my safe. Favorite firearm in that caliber is an OM Ruger Blackhawk I've had since the 60's.

40 S&W (I can hear the gasps.) in my M&P 40c
It comes down to the hatcher rating. While I know there is some really good for 9mm. I like knowing the the 40 S&W packs the perfect balance between knock down power and to much recoil. I also shot 40 S&W well.
A extremely close second is the .44 Magnum in 629. It really come down to the revolver than the round. I like 44 Mag for the BIG power & like .44 special for precision.

1st would be a .22 LR... fun all day and cheap practice.!!!! In way too many different styles of platforms to pick from... but the newest is any one of 4 SW M&P 15-22's
Big bore pistol...can only narrow to 3.!!! First is a .38/357 platform...was and is my first handgun owned...Dan Wesson 15/6VH....second would be my S&W model 29 8 and 3/8" .44 mag...just fun, fun, fun.!!! And daily CCW is a new Glock 30 slimframe in nothing else but the wunnerful .45ACP.!!!!
Shotgun would be 20 gauge...in my Class 3 NFA Ithaca 37 13"... Has a slamfire hammer and I can put all 5 rounds in a 6" pie plate at 10 yards from the hip...
Big rifle...well as the tag name says... .30-06....In an M1 Garand... cant be beaten...versatile, ammo everywhere, game and man and vehicle stopper.!!!!

"Caliber" means cross sectional diameter
.38 Special and .357 Magnum are the same caliber.
Same for .22 LR and .223 Remington.
