As far as I can tell, there is no federal (DOT) law that prohibits having a firearm in a commercial vehicle. You, then, would have to make sure that you are following all state and local ordinances that might modify that. Various employers will also have varying rules regarding this. I guess your safest bet, and probably not what you want to hear, is that you should consult an attorney licensed in the states you intend to travel in.

Somewhere I once read it really boils down to your companys rules.

it should not matter your company rules as far as what the law is should it; company rules should not effect the law I would think.

Nope. It doesn't affect the law.

But, if you work in an ... dang ... I forget the term ("At will employment"?) state, then you could be fired for breaking the rules. I know it's not at all the same as breaking a law, but if you like your job then you could be putting it at risk.

I've driven semis all over the US and Canada while out on tour with different shows. The problem with carrying while drivivng is dealing with the local laws, you may be OK in one state but your license no good in another. Some states consider your vehicle to be covered under the castle law, others don't. Unless you're sure of the laws in each place you drive, you may want to leave the gun at home since a violation of a gun law in a commercial vehicle can cost you your commercial license and the truck with cargo at least temporarily.

jlanecole is correct.
I was one the first fully certified motor carrier safety Troopers in the state of IL in 1983 and I taught motor carrier safety law at our academy for several years.
There is nothing in the MCS laws which prohibits a firearm in the vehicle. The only laws that apply are which ever state the driver is in concerning firearms in vehicles.
You also have to be aware of your company policies. The old rule applies - "boss' truck, boss' rules." Just because something is legal doesn't preclude an owner from prohibiting it. Many times it's the company's insurance carrier that has a say.
