Make sure the yoke sideplate screw is tight. Check the spring/plunger in the yoke screw. Make sure the chambers are fairly clean. I assume you have DS-10 speedloaders, they do require a bit of a push. Holding the gun with 2 fingers thru the cylinder window so you've got hold of the cylinder helps.
