Are there and dimensional differences with newer 627 8-shot cylinders that would prevent fitting with an older 28-2 frame?

Yes. You cannot put an 8 shot cylinder in a 6 shot frame, or a 6 shot cylinder in an 8 shot frame without dire consequences. The reason is that the 8 shot cylinder bolt circle (centerline of chambers) is .06 larger diameter, or .03 larger radius. That means that the 8 shot chamber would be vertically misaligned to the barrel by .03 in a 6 shot frame. If you fired the gun with that much misalignment, it would probably blow it up. I say probably because I'm not going to try it to find out.

Thank you, would you happen to know if the 8 shot cylinder bolt circle would be compatible with a .44 or .45 frame?

Thanks, I didn't know that. I thought an N frame was an N frame. Is a 7 shot L frame the same as a 6 shot? I've always thought it was.
