Any idea what would happen if I accidentally spilled a few drops of locktite in the hole S&W forgot to fill on my new 642?

Yes. if the lock decided to lock up while shooting your gun you would have no way to unlock it!

So if you do this, be sure the lock has been DISABLED first.

Don't do it! Locktite is not like an adhesive. It only sets-up in the absence of air, ie; in-between threads. It would not work in the way that you're hoping for. In addition to that, it could get into the hammer pivot pin, trigger pivot pin, and other parts, gumming up your revolver and making it unreliable.
