The short answer is yes. However, I believe that the purpose of the "third" post is to block the bolt (while the trigger is all the way back during firing) from traveling forward and potentially allowing the cylinder to open during heavy recoil (i.e. magnum). The hands in my parts bin are all "3 post" variety. But, if necessary I would not have a problem using a 2 post hand in 22 rimfire, Model 14, etc.
