Testing for yoke endshake

When testing for yoke endshake, do you reinstall the yoke screw or test without it? There is a video from Power Custom where he is fixing yoke and cylinder endshake, alignment, etc. and he performs the yoke endshake test without the screw installed. He says that the yoke must fit tightly to the frame, so I suppose the screw would be irrelevant in the yoke to frame fit equation. But I see other videos of people testing with the screw installed, and that can tighten things up a bit regardless of yoke to frame fit.

I have a 19-3 which has play with or without the screw, so that obviously needs to be remedied. But, I have a (very lightly used) 29-10 which is solid with the screw installed but has play without the screw. So, does this need to be shimmed as well?
