On some older S&W revolvers there is a screw on the front of the trigger guard. What is the purpose of this screw?

Originally (after 1905) the cylinder stop spring was installed from outside, and accessed by that screw. Around 1960 give or take, they eliminated that extra step and installed the spring under the stop from the inside.

That's true for the K and N frames.
The elimination of the screw was made on the Model of 1953 New I frame and New J frame.
