But if pressed the 38 special for J frame self defense, cost/recoil/accuracy. the 22lr for learning/honing skills for both new & advanced, getting over flinches and back into the zone, a great confidence booster. 45ACP for pure fun & knockdown energy on bowling pins.

.22 LR. Its cheap, accurate, and fun!
With a brick of .22s you can spend all day at the range, practice the fundamentals, don't bother to collect the brass, not wear yourself out and have a darned good time. It don't get no better than that.
I'll give the .38 Special a close second.
This would be my choice also.
.357 magnum..... cus it will do anything I "need" it to do.... and guns chambered for the .357 magnum can also "handle" any .38 special load out there!!! LOL

This is a really tough one for me...for a semi auto I'd go with .45 ACP because I don't have to have the latest in high tech ammo. (I use it in my .45's if available.) the 9x19mm is a very close second.
For revolvers I'd go .38 Special because I can get super light 9 oz J-frames up to (technically) a 47 oz N-frame, although I've been really wanting to try a .44 special.
I just realized I like old and slow!

I'm a big fan of the .357 magnum 125gr JHP from Black Hills. It works well in my handgun and my Henry Big Boy.

In nearly 50 years of shooting, if you asked me at any given time I might have said .38 Special, .45 ACP.
But interspersed in those years was one cartridge that I've always considered way ahead of its time and one that will continue to dominate across most of the world.

If shear volume sent down range is a judge of "favorite handgun round" it would have to be the 158 gr. LSWC in .38 spl. at 850-950 FPS.

Like so many have already said, this is an extremely difficult question to answer. To make it a tad bit easier to answer, for me, I put it into a combination question. If I could only have one handgun, obviously, in one caliber, what would it be.
No doubt in my mind, a revolver, S&W .45 Colt Mountain Gun 4" Barrel. The .45 Colt can be loaded very mild for small game, etc., or it can be loaded pretty hot and can stop, almost anything, that lives in the USA.

For work we carried concealed and were provided with 9 mm Federal Premium LE HST sub sonic 147grain hollow points with a low muzzle flash. Decent penetration, decent ammunition for tactical defense. Recoil is not bad. Retired now and still use it. If they felt it was good round to protect us, for what we needed it for, then I'll keep using it till someone shows me a better alternative.
