I suspect the OP is concerned about the consistency of lockup, not the fact of lockup.
That a barrel locks up and the gun functions does not mean that the slide and barrel will always return to the same starting position with each shot. If these components don't lock up the same with each shot, accuracy will suffer -- because the sights (on the slide) and the barrel won't be pointing to the same points with each shot.
I won't attempt to answer the original poster's question of how best to assess/measure (or improve lockup) as I will only attempt the most rudimentary of gunsmithing tasks on my own -- but I do know the steps needed to assess or improve lockup can vary depending on the gun's design.
