I'm not sure what you mean by fluorescent.
Do you mean self-illuminating sights as in tritium sights, or do you mean photo-luminescent as in glow in the dark watches?
A self illuminating Tritium front sight for a 581 isn't exactly common, or easy to install. You might try one of these if they are available for your 581: XS Sight Systems - Handgun Sights - 24/7 Express
Photoluminescent powder can be mixed with epoxy and applied to your existing front sight but I really haven't see it done, and it would likely make the sights harder to use in daylight.
The best solution for low light shooting is to learn how to use a flashlight with your revolver. LED flashlights have become so small and bright, you can carry a usefull light easier than you can cary an extra speed loader, speed strip, or magazine.

My 581 has a plastic insert in the front metal sight which appears to originally have be a red luminescent material that has turned almost black. When I am at the indoor range I have trouble seeing the sights, so I was thinking I could replace this insert. I guess the other option is to paint the sight with a flourescent paint.

S&W doesn't sell new inserts, since these are cast into the sight.
You either buy an entire new front sight (on pinned-in sight guns) or you cast a new insert.
Any gunsmith can cast in a new insert in about any color you want.
If you're handy with small jobs, you can even do it yourself using epoxy glue and a tiny bit of model airplane paint.
The fastest fix is to just paint the sight with model paint. Fluorescent Red shows up nicely.

Brownells and others sell kits to replace the inserts, or S&W could do it for you.
Between the price of a kit and shipping to and from S&W I think it's a wash cost wize.
The savings with kits really begins to show when doing more than one revolver.
