That pop-up is a scam!!
I suspect it is. OP should clear his browser history/cookies/temporary files and log back in.
Well then MS Defender is not doing a very good job 
I have never used Microsoft Defender.
I have been using both Norton 360 and Spybot -Search & Destroy + AV 2.9 for many many years.
Every 9 days I do a full update and full system scan with both.

What chad said but also run your defender virus scan before you log back in. Make sure it's up to date.

Have you been to gunboards today? That is looking very hacked.
I have a Chromebook so no worries.
You have something hung up in the web page temp files or cookies. The Defender pop-up itself is likely a browser hack.

Happened Yesterday ... the voice kept saying Call the number below and speak to our Technichan ... He will help you ...
Warning Warning Warnung ... do not turn your computer Off and On ... This Action will Destroy your computer you must call the phone number !!!
I was at my daughters house on her computer and when she came home I showed her what was going on ...
She knew how to fix / unlock it ... she turned it OFF ... let the computer do it's reset thing ... and turned it back ON ....
Wah-Lah ... Fixed the problem like Magic !
Don't listen to the voice or call the phone number ...
Do turn the computer Off then wait a min. and turn back ON !
