I've had the cylinder locking lug come loose and get lost on my 29-5 44 mag. Is the forward side plate screw what also holds the cylinder in place and if so, what's the part # and where can I find one?

What is a locking lug?
The locking bolt is pinned in the barrel lug.
The bolt (AKA cylinder release.) is under the hammer and aint goin' nowhere..

If you mean the piece that swings out and holds the cylinder by the extractor rod, that is the yoke. It is held in place by a special scew. The low one on the side plate forward of the trigger. There are two types. Newer ones have spring and plunger in front of scew, older have no plunger or spring and must be fitted to long and they drag on slot in yoke. Is your screw in this location missing. I think I have heard of the new style with spring plunger failing.

There is a "frame lug" located on the frame side of the gun, at the bottom rear of the cylinder window.....it holds the cylinder assembly forward when the cylinder is opened. Is that what you are referring to? Is is pressed in and secured by peening on the older models. (frame lug part#03103)

The number on the front sideplate screw assembly is 22533 (blue)
