I purchased a used 25-5 45 long colt. When I took it to the range to put on paper, I wanted to make a rear sight adjustment. I cannot turn the windage sight either way more than a half turn. I need some advice on the best way to correct this. I assume the adjusting screw has been forced or stripped. Can I replace the sight?

You'll probably have to take the sight apart to see what is causing it to bind. This usually means you'll destroy the screw and nut in disassembly. You'll want to order replacement parts before you start. Watch out for the very small spring and plunger that are contained in the head of the windage screw. They are VERY easily lost. Some people recommend disassembling the sight inside of a plastic bag. I've never done that, but I read that suggestion often. 
Check the nut on the left side of the sight and see if it is moving when you try an adjustment. I've seen some that were not properly installed. Agree on penetrating fluid too.
Heat sight with hair dryer. Spray/drip a good penetrating oil such as Kro-Oil on sight and let it sit overnight. Then try to turn. You could also put screwdriver in slot and tap (LIGHTLY) with a small hammer and try to turn.
Thanks everyone for the information. I tried the Kro-oil trick then tried again which broke the screw. I have ordered parts and a spanner wrench to replace the broken parts. After watching a u-tube video on how to replace the sight it looks pretty straight forward.
