Not a challenge at all: Last Train From Gun Hill.

John Wayne and Clint Eastwood (to a much lesser extent) were in some good westerns, but I can only recall perhaps one or two of the very best that John Wayne was in and can't think of any that Clint Eastwood acted in.

These threads are incredibly subjective; few have the same film tastes.

It's hard to ignore The Searchers. I think it might be the best western ever made. But we are taking John Wayne out of the mix so we can't count it. I'm going to cheat and say the Coen brothers version of True Grit. Jeff Bridges was great in this move. Like everyone else I go back and forth but I think that's my favorite right now. Close behind is The Assassination Of Jesse James By The Coward John Ford. The cinematography and music really give it a moodiness. These are relatively new movies and both seem to have paid a lot of attention to period realism. There is noting more distracting than watching a move that was supposed to take place in the 1870's with everyone carrying 1892 Winchesters.

He was plumb handy at shooting big holes in far away buckets.

That’s an example where whether it is a Western or not is debatable. same with Quigley Down Under. Regardless, Treasure is one of my favorites. I remember that Nixon once said that it was his favorite movie.
